Position Overview
The Sah laboratory at Washington University School of Medicine in St. Louis (Department of Internal Medicine, Cardiology) and the Center for the Investigation of Membrane Excitability Disease (CIMED) focuses on the exploration of Transient Receptor Potential (TRP) and LRRC8a/SWELL1 ion channel dynamics and signaling pathways. Utilizing advanced methodologies such as electrophysiological techniques (including patch-clamp), imaging modalities, genetic mouse models (incorporating CRISPR/cas9 technology), as well as biochemical and molecular strategies, the laboratory investigates the regulatory mechanisms of novel ion channels, specifically TRP/LRRC8 channels, in various tissues including cardiac muscle, skeletal muscle, adipose tissue, endothelium, and pancreatic islet cells.

The primary objective of the Sah laboratory is to elucidate the influence of ion channels and mechanosensing on cellular and organismal metabolism. Additionally, we integrate medicinal chemistry with cryo-EM structural analysis, molecular docking, and functional assessments to facilitate the development of innovative ion channel modulators aimed at addressing human health challenges.
