St Louis, Missouri, United States Washington University in St. Louis Full time
Job Description

The primary objective of this role is to engage in life sciences research activities under the guidance of the Principal Investigator (PI). This position will broadly encompass the investigation of leukocyte movement within the context of tumor immunology.

Utilizing in vivo tumor models, we examine the impact of altering chemokines on leukocyte migration in the tumor microenvironment. Our laboratory focuses on the molecular and cellular processes associated with leukocyte trafficking in this environment, aiming to develop innovative immunotherapeutics that can be swiftly and effectively translated into clinical applications. We are pioneering various delivery methods (such as nanoparticles, viruses, and fusion proteins) to direct specific chemoattractants to tumor sites, employing these strategies individually and in combination with other immunomodulatory techniques.

Another significant area of focus for our lab is translational immunotherapy in prostate cancer. We are collaborating closely with leading experts on the initial trial of neoantigen vaccines for prostate cancer patients. Our lab will conduct thorough analyses of all patient samples collected during this trial to assess immune responses. Additional projects involve studying patient tumor samples to gain insights into the tumor microenvironment and responses to immunotherapy. In partnership with other research teams, we are utilizing ctDNA, NGS, and single-cell RNA sequencing to explore the immune landscape of prostate cancer and identify predictive biomarkers for therapeutic response. Our unique prostate cancer tissue bank contains hundreds of tumor samples, matched plasma, and bone marrow aspirates, and we will employ techniques such as CyTOF to investigate these tissues.

Key competencies for this role include the ability to acquire new techniques and proactively advance projects. Ideal candidates will possess some experience in the following areas, although not mandatory: molecular biology, tissue culture techniques, small animal handling, tumor models, and various immunoassays (e.g., ELISAs, Luminex, cytotoxicity assays), as well as multi-color flow cytometry and FACS data analysis. Other responsibilities will include analyzing human tissues using techniques such as qPCR, Western blotting, multiplexed IHC/IF, and single-cell RNA sequencing. Candidates should be capable of analyzing data and drafting initial reports in both manuscript and grant formats. Participation in weekly lab meetings and larger group discussions as part of our Center for Human Immunology and Immunotherapy Programs (CHiiPs) is expected. Ultimately, candidates should aim to present their findings at national conferences and work towards securing their own grant funding.

Required Qualifications:

  • BS/MS in a relevant scientific discipline
  • Strong organizational skills to assist with laboratory management tasks (e.g., protocol development, ordering supplies)
  • Ability to work independently and take initiative in advancing projects
  • Some experience with techniques such as tissue culture, molecular biology, and flow cytometry
  • General computer proficiency and the ability to quickly learn and master analytical software
  • Excellent analytical skills and sound judgment
  • Capability to work under deadlines with general guidance is essential
