Position Overview
The Sah laboratory at Washington University School of Medicine in St. Louis focuses on the exploration of mechanisms governing Transient Receptor Potential (TRP) and LRRC8a/SWELL1 ion channel regulation and signal transduction. Utilizing a range of methodologies including electrophysiological techniques (such as patch-clamp), imaging modalities, genetic mouse models (incorporating CRISPR/cas9 technology), as well as biochemical and molecular strategies, the laboratory investigates the intricate regulation of signal transduction pathways involving novel ion channels, particularly TRP/LRRC8 channels, across various tissues including cardiac muscle, skeletal muscle, adipose tissue, endothelium, and pancreatic islet cells.
The primary objective of the Sah laboratory is to elucidate how ion channels and mechanosensitive signaling pathways impact cellular and organismal metabolism. Additionally, we integrate medicinal chemistry with cryo-EM structural analysis, molecular docking, and functional assessments to inform the development of innovative ion channel modulators aimed at addressing human health challenges.
