Principal Scientist / Associate Director – Biotherapeutics Engineering

Xilio Therapeutics is a pioneering biotechnology firm focused on the discovery and advancement of tumor-activated immuno-oncology therapies. Our mission is to enhance the quality of life for cancer patients by minimizing the systemic side effects associated with conventional immuno-oncology treatments. Utilizing our proprietary platform, we are advancing a pipeline of innovative, tumor-activated therapeutic agents designed to maximize their efficacy within the tumor microenvironment, including tumor-activated cytokines, antibodies (including bispecifics), and immune cell engagers.

POSITION OVERVIEW:

We are in search of a highly qualified and seasoned Principal Scientist or Associate Director in Biotherapeutics Engineering. The ideal candidate will be responsible for the design and optimization of biologics-based therapies, leveraging expertise in structural biology alongside cutting-edge computational methodologies. A robust background in structural biology, protein engineering, and antibody discovery processes is essential. The candidate should also possess experience in applying machine learning techniques and molecular modeling to enhance the biophysical and biological characteristics of biotherapeutics. Leadership experience in managing cross-functional teams and driving projects from conceptualization to in vivo proof of concept is highly valued. If the candidate has a strong interest in team management, the role may be elevated to Associate Director, overseeing a small team.

Key Responsibilities:

  • Utilize advanced in silico biotherapeutic engineering tools, such as CCG-MOE or Schrodinger-Maestro, to design and refine biologics-based therapies.
  • Implement state-of-the-art generative AI, machine learning, and molecular modeling techniques to discover novel binders and enhance the properties of biologics, focusing on target engagement, developability, immunogenicity, and protease specificity.
  • Conduct biochemical and biophysical analyses of engineered proteins to assess functionality and establish structure-function relationships.
  • Lead or co-lead research project teams, responsible for defining strategic objectives and ensuring effective execution to meet project milestones.
  • Represent the Protein Engineering discipline in project team discussions.
  • Assist in the preparation and documentation of key experimental findings for regulatory submissions.
  • Maintain detailed experimental records in electronic lab notebooks (ELN).

Qualifications and Skills:

  • PhD in Structural Biology, Chemical and Biomolecular Engineering, or a related discipline, with a minimum of 7-9 years of relevant post-doctoral experience in either industry or academia.
  • Extensive expertise in biotherapeutics engineering, particularly in computational drug design and optimization.
  • Proficiency in protein design and bioinformatics, including molecular modeling techniques (homology modeling and molecular dynamics simulations) using tools such as MOE, Maestro, NAMD, Schroedinger, or VMD.
  • Strong analytical and problem-solving capabilities for the computational analysis of protein, DNA, and mRNA sequences.
  • Experience in applying machine learning methodologies to enhance the properties of biologics is preferred.
  • Familiarity with various generative AI/machine learning algorithms and experience in data mining through APIs is advantageous.
  • Knowledge of cloud computing and coding skills (SQL, R, Python) for data analysis and management is desirable.
  • Experience with electronic lab notebooks (e.g., Benchling) for data management and analysis is preferred.
  • Proven experience in supervising team members or managing CRO projects is highly regarded.
  • A demonstrated history of leading cross-functional teams and successfully delivering projects from concept to proof of concept is preferred.
  • Exceptional leadership and communication skills to effectively guide cross-functional teams and collaborate with various stakeholders.
