Position Summary


Affinia Therapeutics is seeking an experienced scientist (job level will be commensurate with experience) to join our Vector Biology laboratory team.

You will be part of a team dedicated to advancing AAV-based gene therapies using your expertise in Next Generation Sequencing (NGS), molecular biology/cloning, and cell-based assays.

This is a lab-based, onsite role where you will actively contribute to capsid discovery and disease programs.

The role is located at our state-of-the-art facility, in Waltham, MA, and reports to the Head of Vector Biology.

Primary Responsibilities

Be an integral member of our NGS platform, receiving and processing samples for sequencing using our Illumina and Nanopore devices

Create and execute cloning strategies to generate constructs that express Therapeutic Cargos such as proteins and artificial microRNAs

Independently perform cell-based assays to determine AAV functionality

Work cross-functionally with our Computational Biology and Process Science teams in these efforts

Write, prepare and present technical data, technical reports and standard operating procedures (SOPs) for internal use and tech transfer

Maintain an up-to-date electronic notebook, participate in meetings, prepare presentations and communicate to leadership as needed

Minimum Qualifications

4+ years of industry experience

Hands-on experience with techniques required to extract RNA/DNA from virus, mouse, non-human primate and mammalian cell culture models, and then process for NGS using Illumina Novaseq (amplicon-based) and Nanopore MinION devices

Hands-on experience with molecular biology techniques including plasmid cloning, DNA sequencing analysis, plasmid preparation, restriction digests, agarose gel electrophoresis, Gibson assembly

Hands-on experience with cell-based assays including cell culture and cell transfection/transduction, fluorescence-based readouts using plate readers or FACS, RNA/DNA extractions and qPCR/ddPCR, protein gels and Western blots

Excellent written and oral communications skills, including experience writing laboratory SOPs, protocols, and reports

Ability to organize and communicate scientific data to management and internal stakeholders

Ability to work independently to determine and acquire new methods and scientific knowledge

Strong collaboration skills

Prior AAV-related experience a plus

Ability to thrive in a fast-paced environment flexibly and collaboratively
