Position Summary
We are seeking a driven Scientist in Protein Engineering to enhance CAR T design and optimization, working collaboratively with both internal teams and external partners. The primary focus of this role will be to refine novel mRNA-based CAR T constructs for in vivo applications across various therapeutic areas.
Key Responsibilities:
- Lead the discovery of product candidates by devising and implementing detailed research strategies.
- Engage in all facets of antibody discovery and receptor engineering, including library design, screening, and binder identification to achieve program objectives.
- Develop CARs, multi-format antibody constructs, and other protein-based entities for both cell-based and recombinant expression.
- Formulate molecular biology strategies for cloning relevant constructs and constructing libraries.
- Create plasmids and lentivirus vectors.
- Employ advanced techniques such as electroporation, viral delivery, and nanoparticle transfections to modify mammalian cells.
- Utilize flow cytometry and cell-based functional assays to thoroughly characterize mammalian cells.
- Perform meticulous mammalian cell culture procedures adhering to aseptic techniques.
- Ensure compliance with stringent laboratory standards, including detailed documentation in an online laboratory notebook, instrument validation, and personal safety protocols.
- Build collaborative relationships within a cross-functional team environment, acting as a key contributor to project teams focused on developing, refining, and standardizing cell-based assays across multiple initiatives.
- Effectively organize and communicate data through written and verbal channels, actively participating in project team discussions.
Qualifications:
- Ph.D. in a relevant scientific discipline (preferably Protein Engineering, Immunology, Cell Biology, or related fields) with a minimum of 2 years of industry experience or equivalent.
- Expertise in structure-based engineering of CARs, antibodies, and/or other proteins.
- Proficient in molecular biology techniques, including Gibson, Golden Gate, and library construction methodologies.
- Strong background in mammalian display and NGS methodologies for antibody discovery and CAR-T engineering is highly desirable.
- Prior experience in the development of antibody therapeutics, particularly bispecific or ADC, is advantageous.
- Demonstrated success in producing lentivirus/retrovirus and engineering primary T cells for CAR or TCR production.
- Familiarity with non-viral gene transfer techniques for mammalian cell engineering is a plus.
- Experience in designing and executing assays to assess T cell functionality, including evaluations of cytotoxicity, activation, proliferation, and cytokine production.
- Knowledge of plate-based assay development and proficiency in automated liquid handling systems is beneficial.
- Familiarity with scientific data analysis software such as GraphPad Prism, FlowJo, or CellEngine, and experience analyzing large datasets using R, Python, or other computational tools is advantageous.
- Proven ability to conceive and execute research projects independently while contributing to team efforts.
- Strong organizational and time-management skills, with the ability to manage multiple projects effectively and maintain thorough documentation practices.
- Excellent communication and interpersonal skills, with a strong team-oriented mindset.
- Capacity to be adaptable, responsive, and highly organized with meticulous attention to detail.
