(Senior) Scientist, Lead Discovery

Site Therapeutics aims to map—down to the nucleotide—the genetic determinants driving immune cell function in human health and disease and to use that information to develop first-in-class small molecule therapeutics for autoimmune disease. Leveraging technology developed in the Marson Lab at UCSF/Gladstone, we will conduct high-throughput functional genomics in human cells to build a blueprint for fine-tuned control of immune cell function. This detailed map will lead to the development of new therapeutics across modalities for a range of immune-related conditions. We are backed by The Column Group, a leading life sciences venture capital firm.
We are seeking a highly motivated and innovative biochemist and small molecule discovery scientist to join our growing Lead Discovery group at Site Tx. The Lead Discovery group drives the translation of novel functional sites on proteins, identified by our base editing platform, into small molecule effectors that phenocopy these functional effects and enable the development of small molecule therapeutics. In this role, you will provide expertise in devising and developing the functional assays and screening methodologies that will identify these molecules and characterize them biophysically and structurally to enable progression through structure-based drug design campaigns. This role represents an opportunity to join a company at the earliest stages and have an outsized impact in its growth and success.
Primary Responsibilities
- Using biophysics, biochemistry, and structural biology to characterize and elucidate the function of novel protein target sites identified by Site Tx’s base-editing functional genomics platform
- Operationalizing and conducting small molecule discovery on these target sites by various approaches, including biochemical high-throughput screening and DNA-encoded library (DEL) selection
- Characterizing hit compound mechanisms of action through biochemical, biophysical, and structural characterization
- Progressing these small molecules toward the clinic through structure-based drug design campaigns as a member of a cross-functional team consisting of medicinal chemists, computational chemists, cell biologists, and pharmacologists
- Presentation of research findings to senior management and other stakeholders, both within and outside the company
Qualifications:
- Ph.D. in biochemistry, biophysics, structural biology or related fields with 2+ years post degree experience
- Demonstrated success using screening methodologies and SBDD to identify, characterize, and progress small molecules against novel therapeutic targets
- Experience using protein science to develop high quality, functional protein reagents for downstream assay development and discovery (multiple expression systems, protein purification, QC, optimization)
- Experience developing fluorescence-based activity and protein interaction assays (TR-FRET, FP, etc.) for the characterization of protein-protein and protein-small molecule interactions
- Experience developing biophysical protein interaction systems (SPR, GCI)
- Strong track record of scientific accomplishment (publications, presentations, industry experience)
- Strong desire to operate and thrive in a fast-paced, collaborative, and exciting scientific environment translating novel biological insights into transformative therapeutics for patients
