The laboratory of Dr. Sattar Khoshkhoo in the Department of Neurology at Brigham and Women's Hospital and Harvard Medical School is seeking highly motivated postdoctoral fellows with a passion for studying the genetic and neurobiological mechanisms of neuropsychiatric diseases including epilepsy. The positions offer a minimal annual salary of $70,000 and are adjustable based on research experience.

The mission of the Khoshkhoo Lab it to examine the contribution of somatic genetic variation in healthy human brain development and in neurologic diseases. Our work spans the direct investigation of the human brain tissue to using cells lines and model systems. Our aim is to identify novel somatic mutations in the healthy and diseased human brain, to examine their developmental origins and functional impact at the level of individual cells and neural circuits, and to find new drugs and targets that could be leveraged for the development of targeted and disease-modifying therapies. Dr. Khoshkhoo and his team recently discovered that ultra-rare pathogenic somatic single nucleotide variants are present in >40% of the surgically resected hippocampi from epilepsy surgery. Interestingly they also showed that extremely rare pathogenic somatic variants could be found in neurotypical control brains. Our current research directions that are relevant to this position include 1) Identifying novel forms of somatic genetic variation in the healthy and diseased human brains using new sequencing approaches such as duplex and long read sequencing, 2) Lineage tracing to examine the contribution of hippocampal neurogenesis to the development of somatic mutations using single cell whole genome sequencing and other experimental approaches, 3) Understanding the cell type of origin and pathogenic mechanisms of somatic mutations in neurologic diseases using novel single cell and spatial transcriptomics and epigenomics, iPSC-derived co-culture models, and mouse experimental frameworks using neurophysiology and functional imaging/perturbations, 4) Investigating the mechanisms underlying selective enrichment of oncogenic somatic mutations in non-neoplastic neurological diseases, as well as their functional impact on the transcriptome and epigenome of carrier and wildtype cells. The Khoshkhoo lab has one of the largest collections of tissue from epilepsy surgery and actively and regularly collaborates with other labs at Harvard and around the world to engage in productive, highly collaborative, and impactful science that will ultimately help advance human health. Candidate qualifications include:

  • PhD and/or MD in genetics/genomics, molecular biology, neuroscience, proteomics/biochemistry, or other related fields.
  • Demonstrated expertise in one or several of the following areas:
    • Human genetics including the experimental and analytical aspects of next generation sequencing.
    • Single cell genomics and/or spatial biology.
    • IPS-based model systems for neuroscience applications including neuronal 2D and 3D cultures.
    • In vivo neurophysiology and functional imaging including calcium/voltage imaging.
    • Gene editing including high-throughput CRISPR screens, prime/base editing.
  • Proficiency in basic molecular biology including cloning, cell culture, immunostaining, and microscopy.
  • Strong quantitative and analytical skills, and familiarity with one or multiple programming languages including R/python/MATLAB.
  • Track record of first-author original research papers.
  • Strong oral and written communication skills.
