Position Overview:

The Senior Staff Scientist role is available within the FIB-SEM Collaboration Core (F-SCC) at Yale School of Medicine. This position is pivotal in advancing volume Electron Microscopy, leveraging cutting-edge Focused Ion Beam Scanning Electron Microscopy (FIB-SEM) technology for extensive imaging with nanometer isotropic resolution. Reporting to the Director of F-SCC, the selected candidate will take on a leadership position in overseeing the electron microscopy (EM) sample preparation laboratory within F-SCC.

Key Responsibilities:

The candidate will be tasked with enhancing current EM sample preparation protocols for biological specimens and innovating new methodologies for clinical samples. Responsibilities include:

  1. Executing EM sample preparation requests across a variety of biological and human samples, utilizing the reduced Osmium Tetroxide-Thiocarbohydrazide-Osmium (rOTO) protocols to ensure optimal sample quality in terms of ultrastructure preservation, membrane contrast, and resin infiltration.
  2. Conducting transmission electron microscopy (TEM) thin section preparation and imaging to assess sample quality.
  3. Optimizing existing EM sample preparation protocols to enhance ultrastructure preservation and sample contrast.
  4. Establishing transcardial perfusion operations on animal models.
  5. Developing innovative methods for clinical specimen preparation.
  6. Creating an automated EM sample preparation pipeline.
  7. Performing FIB-SEM sample preparation, which encompasses sample mounting and precise targeting of specific regions of interest using ultramicrotome guidance and x-ray micro-CT.

Essential Duties:

  1. Conducting a wide array of specialized scientific techniques and procedures in the laboratory.
  2. Managing resource planning, allocation, and laboratory renovations, while establishing and implementing the operational framework aligned with long-term objectives.
  3. Staying updated on emerging technologies, instrumentation, and methodologies, including procurement of new instruments.
  4. Developing, analyzing, interpreting, and summarizing scientific findings.
  5. Establishing policies and procedures governing laboratory operations.
  6. Independently addressing and resolving biotechnological challenges through research and comprehensive literature reviews.
  7. Determining when additional analyses are necessary, coordinating those analyses, and interpreting the resulting data to implement tailored protocols.
  8. Authorizing and organizing laboratory and equipment usage to ensure timely project completion.
  9. Performing additional duties as assigned.

Qualifications:

A Master's Degree in chemistry, biology, biochemistry, or a related field, accompanied by five years of relevant experience, or an equivalent combination of education and experience is required.

Required Skills:

  1. Proficiency in developing new EM sample preparation protocols and implementing automated pipelines to support advancements in volume electron microscopy.
  2. Ability to research and interpret scientific literature, protocols, and standard operating procedures.
  3. Experience in operating and modifying laboratory instruments for enhanced performance and troubleshooting.
  4. Capacity to anticipate project requirements and strategically plan resource allocation for successful project execution.
  5. Skill in maintaining laboratory facilities and adapting to new procedures as needed.

Preferred Qualifications:

A Ph.D. in biology, chemistry, biochemistry, or a related discipline, along with five years of pertinent experience, is preferred. Candidates with a strong background in EM sample preparation, a keen interest in automation, and certification as an Electron Microscopy Technologist will be given special consideration.

Work Schedule:

Occasional weekend and evening hours may be required.
