Description
Our group works on bone physiology. We study both bone resorbing (osteoclasts) and bone forming cells (osteoblasts). We primarily use animal models to study bone by micro-computed tomography analysis of bone structure, histology, serum markers and primary cell culture. Working under general supervision of the Principal Investigators and in accordance with established hospital policies and procedures, this individual will assist with mouse colony management, quantitative PCR and other molecular biology techniques, bone analysis with CT and histology and primary cell isolation and ex vivo culture. This individual will be responsible for detailed record keeping of both text and digital images.
We are a small and supportive group which makes this a great learning opportunity for a new graduate. We also work closely with several other groups and have a joint lab meeting such that the work community is relatively large and diverse and exposure to a wide variety of scientific topics is assured.
PRINCIPAL DUTIES AND RESPONSIBILITIES:
Maintains breeding colonies of various mouse strains. Sets up breeding cages, prepares material for genotyping, performs PCR and interprets results to determine genotypes. Maintains accurate records of all breedings. Designs and executes research experiments, interprets results, and troubleshoots techniques required to complete experiments with help of supervisor. Experiments may include any of the following: Isolates bones and performs histologic or micro-computed tomographic analysis using standard techniques. Image generation and annotation.Data analysis using Graph Pad Prism.Performs ELISA assays on mouse serum and/or cell culture supernatants. Isolates primary cells and maintains them in short-term cell culture. Assays cellular function utilizing a variety of standard in vitro assays. Isolates primary cells and performs flow cytometric analysis and/or prepares cells for fluorescence activated cell sorting.Isolates organs or primary cells, prepares RNA and performs real-time PCR on samples. Lab maintenance responsibilities include preparing media, maintaining cell culture room, and preparation of various stock solutions. Performs all other duties and responsibilities as directed.
Qualifications
. in a biologic/physical science is required; prior laboratory experience outside of coursework is a plus.
SKILLS/ ABILITIES/ COMPETENCIES REQUIRED:
Broad knowledge of scientific principles, biologic structures, and laboratory techniques, typically acquired through a Bachelor’s degree in biological sciences but interested chemistry or engineering students are welcome. Some familiarity with basic laboratory instruments and scientific procedures. A high degree of computer literacy is a plus. Self-motivation, judgment and the ability to identify potential problems and trouble-shoot problems as necessary. Strong organizational and communications skills, and time management skills are essential for success in this position. Previous experience working in a research lab (3-6 months) or laboratory-based course work are a plus. A one year commitment (through to fall of 2018) is needed.
WORKING CONDITIONS:
Lab environment, about 70% bench and 30% office.
