ABOUT THE OPPORTUNITY:
Valora is building a Protein Science team and is seeking highly motivated and experienced Protein Scientists/Engineers to play a key role in developing next-generation antibody therapeutics. This role offers the opportunity to:
- Innovate in Protein Engineering & Biophysics: Shape the future of our protein engineering efforts and guide groundbreaking discoveries using Valora's proprietary chimeric bispecific antibody platform.
- Impactful Role: Play a key role in developing next-generation antibody therapeutics using our proprietary chimeric antibody platform.
o Design and engineer novel chimeric antibodies using Valora's proprietary platform.
o Develop and optimize methods for protein production and purification.
o Perform analytical and biophysical characterization of antibodies.
o Evaluate the pharmacological properties of engineered antibodies in collaboration with other R&D teams.
o Pay key roles in novel antibody discovery campaigns including antigen generation, and antibody discovery and selection.
o Contribute to IND-enabling studies and IND filings.
o Participate in development of intellectual property applications.
o Mentor junior team members and share your expertise.
o Partner with external collaborators to accelerate drug discovery and technology development.
ABOUT YOU:
- You offer experience: you hold a Ph.D. with a minimum of 10+ years of experience in antibody discovery and characterization in the academic or biopharmaceutical industry (more years’ experience expected for higher level and salary range). Bonus experience: You have experience in supporting R&D therapeutic programs from early-stage discovery to the clinic.
· You are a subject matter expert: you have extensive expertise within the discipline of antibody discovery and engineering. Maintain broad knowledge of state-of-the-art principles and theories and remain current with changes in the field and in the industry. You have strong knowledge of oncology and immunology.
- You are a technical expert: possess extensive hands-on experience in some of these areas:
- Antibody Expression, Purification, Characterization:
§ Expression: DNA manipulation and molecular cloning. Transient mammalian expression and protein purification. Knowledge of protein structure-function relationships and tools for protein modification and expression.
§ Purification: Experience in purifying recombinant proteins to high homogeneity for immunization purposes. Techniques like chromatography and affinity purification are essential (e.g. affinity, IEX, HIC, SEC).
§ Characterization: Experience in protein characterization and QC (analytical SEC, CE-SDS, SDS-PAGE, Western Blotting, endotoxin assays) including biophysical techniques (SPR, Octet, ITC, CD, SEC-MALS).
- Data analysis: Proficiency in analyzing and interpreting data from immunoassays and antibody discovery platforms.
§ Problem-solving and critical thinking: Ability to troubleshoot challenges encountered during antigen design, immunization, and antibody discovery processes.
§ Excellent communication and collaboration: Skills to effectively communicate research findings and collaborate with colleagues from diverse scientific backgrounds.
- Antigen Expertise:
§ Protein design and engineering: Ability to design and express recombinant proteins for use as antigens. This includes knowledge of protein structure-function relationships and tools for protein modification.
§ In silico antigen selection: Skill in using computational tools to identify and prioritize potential immunogenic regions of target proteins.
§ Protein purification techniques: Experience in purifying recombinant proteins to high homogeneity for immunization purposes. Techniques like chromatography and affinity purification are essential.
- Immunization Strategies:
§ Immunogen conjugation: Understanding of how to conjugate antigens to carrier molecules to enhance immunogenicity.
§ Animal models: Experience working with various animal models for antibody production, including understanding their strengths and limitations.
§ Adjuvant selection: Knowledge of different adjuvants and their impact on the immune response and antibody generation.
- Antibody Discovery Techniques:
§ Phage display: Experience in using phage display libraries for antibody selection against target antigens.
§ Hybridoma technology: Understanding of hybridoma generation and subsequent hybridoma screening assays for antibody discovery.
§ ScFv and single-domain antibody selection: Familiarity with techniques for isolating and characterizing single-domain antibodies.
§ High-throughput screening (HTS) platforms: Skill in using HTS platforms for rapid identification of antibodies with desired properties like affinity and specificity.
- Bonus Skills:
§ Experience with next-generation sequencing (NGS) for antibody repertoire analysis.
§ Knowledge of antibody engineering techniques for antibody optimization, generation of bispecific antibodies or antibody drug conjugates.
§ Experience with bioinformatics tools for antibody sequence analysis.
