Dyne Therapeutics is a clinical-stage muscle disease company focused on advancing innovative life-transforming therapeutics for people living with genetically driven diseases. With its proprietary FORCE™ platform, Dyne is developing modern oligonucleotide therapeutics that are designed to overcome limitations in delivery to muscle tissue. Dyne has a broad pipeline for serious muscle diseases, including clinical programs for myotonic dystrophy type 1 (DM1) and Duchenne muscular dystrophy (DMD), and a preclinical program for facioscapulohumeral muscular dystrophy (FSHD).

Role Summary:

As a member of the Platform Oligonucleotide innovation team, the successful candidate will contribute to the development of Dyne’s oligonucleotide chemistry platform and enable the discovery of novel nucleic acid therapeutics. This individual will be responsible for the design, synthesis, purification, and analysis of oligonucleotides and their conjugates. The successful candidate should have technical expertise in modified RNA synthesis and experience with key biochemical assays to measure RNA stability and mode of action.

This role is based in Waltham, MA without the possibility of being a remote role. Applicants must be able to relocate to the area.

Primary Responsibilities Include:

Perform synthesis of oligonucleotides with diverse sugar and backbone modifications at various scales Troubleshoot and optimize synthesis methods to produce high quality oligonucleotides of various lengths Interpretation of analytical data (HPLC, LCMS) to enable improved purity and yields of in-house synthesized oligonucleotides Carry out purification (anion exchange, reverse phase), analysis (IP/RP, IEX, SEC), characterization (LCMS, UV, melting temperature) of oligonucleotides and their conjugates Perform standard biochemical assays to determine stability and mechanism of action of oligonucleotides Contribute both technically and rationally to the hit-to-lead optimization process Cross-functional collaboration to define deliverables and timelines Multi-task across several projects and manage time effectively to achieve results Collect, present, and discuss data at internal team meetings

Education and Skills Requirements:

PhD in chemistry or biochemistry or MSc with at least 3-5 years of industrial experience in the biotechnology industry, or BSc with 5+ years experience Prior experience with the operation of oligonucleotide synthesis and purification equipment is highly valued Familiarity with biochemical assays (RNA stability, gel shift assays, DNA/RNA cleavage assays) is preferred Experience with enzymatic reactions (RNA ligation, IVT, PCR) is preferred Collaborative team player
