Advertised Job Summary:

The postdoctoral fellow will be responsible for analyzing biosynthetic genes, identifying and isolating natural products, determining the chemical structures of the natural products and elucidating the corresponding biosynthetic pathways. The postdoctoral fellow will also be responsible for data analysis, preparation of written reports and manuscripts, as well as mentoring graduate students in the laboratory.

Required Education and Experience:

Requires a Doctoral (Ph.D.) degree in area of specialty.

Preferred Qualifications:

The candidate should have a Ph.D. degree in chemistry, biochemistry or related field by the start date of employment with expertise in natural products chemistry, genome mining, or biosynthesis. This expertise could have been gained during the normal course of a doctoral degree program.

Knowledge/Skills/Abilities:

Natural products chemistry is required; natural products metabolomics, biosynthesis and biological evaluation would be a plus.

Job Duty:

Performs microbial genome mining, biosynthetic gene cluster cloning/capture and expression. Exploring microbial genomes to find genes or biosynthetic gene clusters (BGCs) that have a particular function, often related to the biosynthesis of natural products like antibiotics or other metabolites, using bioinformatic tools. After identification, the candidate will clone these genes or BGCs, typically using techniques like PCR amplification, TAR cloning, and vector-based transformation or conjugation, into more manageable host organisms, such as E. coli or Streptomyces to produce target compounds. The expression condition will be optimized as necessary to efficiently harness the novel compounds they produce, paving the way for potential applications in medicine.

Performs compounds isolation and structure elucidation.Isolating the novel natural products produced by the BGCs using a combination of various chromatography systems such as LH-20, C18 silica gel, and HPLC. Upon isolation of the compounds, the candidate will use NMR, UV, IR, HRMS, and x-ray diffraction to elucidate the chemical structures of these compounds.

Performs biosynthetic pathway and enzymology study. Investigating biosynthetic pathways - sequences of enzymatic reactions that lead to the synthesis of complex molecules from simpler precursors. The candidate will express and purify enzymes involved in the biosynthetic pathway, identify intermediates, determine the kinetic parameters of enzymes, further illuminate the enzyme's specificity and mechanism.

Job Duty:

Assists with biological/physiological study of the compounds discovered. Facilitating a multifaceted exploration into the interactions, impacts, and implications of novel compounds. The candidate will conduct the in vitro assessment of the compounds, often utilizing cell lines to ascertain biological activities, such as potential cytotoxicity, cell proliferation, or specific biomarker modulation. Colonization of mice with certain microbe may also be performed, followed by analysis of a variety of biomarkers and/or phenotypes.

Work with the PI to write manuscripts and reports on funded projects.The candidate will lead the writing of journal articles and intellectual property disclosures emerging from research activities. It is expected that the successful candidate will be experienced in the preparation of publication-quality research artwork and narrative descriptions of results and background. The candidate will work with and receive feedback from the PI to continue to develop their expertise in the field.
