Job Description:
The job involves a critical role in a multi-year research program aimed at discovering fundamental mechanisms that regulate anti-tumor immunity within head and neck cancer. The Research Associate Scientist will perform experiments using animal models and clinical tissues obtained from clinical trials to uncover how tumor-fighting immune cells arrive at and function within tumors. Responsibilities include developing and implementing protocols for processing clinical trial and laboratory specimens, cataloging and tracking human clinical and laboratory specimens using Freezerworks, and preparing single cell suspensions from blood and tissue samples. 

The role also involves extracting and culturing T-cells, conducting single cell transcriptome sequencing, flow cytometric analysis, and functional studies like Elispot and cytotoxicity assays. Additional duties include performing multispectral immunofluorescence microscopy, multicolor flow cytometric assays, and animal studies involving various techniques such as tumor cell transplantation and immune cell administration. This position requires maintaining accurate laboratory records and adhering to BSL-2 protocols for handling human blood, tissues, and cell lines, including HPV-positive samples. The Research Associate Scientist will work closely with the Principal Investigator to interpret and report results, contributing to publications and advancing our understanding of anti-tumor immunity and cancer therapy development.

Experience Required:

  • Ph.D. degree in a scientific field. 
  • Three (3) years of specialized experience plus a Master’s degree is equivalent to a Ph.D.
  • Five (5) years of specialized experience plus a Bachelor’s degree is equivalent to a Ph.D.
  • Minimum of Six (6) years of post-doctoral experience in a cancer immunology laboratory.
  • Experience independently developing, troubleshooting, and performing immunofluorescence and use of multiplex techniques to multicolor analysis of fixed tissues.
  • Experience independently developing, troubleshooting, and performing fluorescence cytomtry using multicolor analysis.
  • Experience independently developing, trouble shooting and preparing cells from tissues, blood, or cell lines, such as for T cell Elispot and cytotoxicity assays, and single cell sequencing.

Requirements:

  • Develop and implement protocols to receive and process multiple clinical trial and laboratory specimens weekly from clinical and animal protocols.
  • Develop customized programs for cataloging, storing, and tracking use and disposal of valuable human clinical and laboratory specimens in multiple ultralow and liquid nitrogen freezers utilizing Freezerworks. 
  • Provide annual reports of all human specimens as required.
  • Develop and perform protocols for processing of blood and tissue collected from clinical trials into single cell suspensions.
  • Develop and troubleshoot protocols to extract T-cells and other tumor cell populations from patient samples for single cell transcriptome sequencing, flow cytometric analysis, and functional studies such as Elispot and cytotoxicity assays.
  • Develop, troubleshoot, and standardize protocols to culture and expand extracted T-cells.
  • Develop and perform new molecular assays on human clinical trial specimens using new platforms, such as single cell sequencing.
  • Develop, troubleshoot, and perform multispectral immunofluorescence microscopy assays on human and murine specimens.
  • Develop and perform multicolor flow cytometric assays on human clinical trial specimens.
  • Design experiments and perform animal (murine) studies involving animal handling, tumor cell transplantation, immune cell depletion or administration, tumor measurements, and standardize treatment techniques such as intraperitoneal injections, IV tail vein injections and oral gavage. 
  • Develop, troubleshoot, and perform sterile cell culture techniques such as maintaining and expanding immortalized cell lines.
  • Ability to maintain accurate and detailed laboratory records.
  • Maintain universal precautions and follow SOPs for BSL-2 handling and analyses of human blood, tissues, and cell lines, including human papilloma virus positive tissues and cell lines.

Preferred Skills:

  • Training in customizing and using Freezer works and maintaining a tissue biorepository.
  • Strong communication skills, both oral and written.
  • Excellent analytical, organizational, interpersonal and time management skills.

Educational Qualifications: Ph.D. degree in a scientific field.
